چکیده

By J. PHILIP SAVITSKY, M.D. C HANGES in platelet adhesivemiess have been noted during the course of many different clinical conditions.t4 Imicreased platelet adhesiveness has been correlated by some investigators with a predisposition to thromboembolic phemiomena and vascular disease,2 but other work has not confirmed this suiggestion.3 It has been show’mi that there is a very close relation between the level of the platelet adhesiveness amid the rate of imsit.iation of clot retraction as measured by the clot retraction time.5 The number and morphology of the platelets and the extent of clot retraction ‘ere found to be unrelated to the platelet adhesiveness and the clot retraction time.5 These observations suggested that there might be a substance outside the platelets which makes them adhesive and accelerates the initiation of clot retractioui. Wright has suggested that fibrin, which is constantly being formed imitravascularly, coats the platelets, making them more adhesive.6 No evidence, how’ever, for this hypothesis has been offered and it has been observed that the fibrinogen level has no relation to platelet adhesiveness or clot retraction time.5 This present investigation attempts t.o demonstrate the existence of a factor circulating in the blood outside the platelets which acts to increase platelet adhesiveness and accelerate clot retraction. Some of the properties of this factor are studied and their relationship to the “viscous metamorphosis” of the platelets, described by Wright and Minot in 1917, is clarified.7

Control of radiation hemorrhage with splenic extracts.

It has been recently observed that prior to the development of the thrombocytopenia in the irradiated dog there is a change in the surface properties of the platelets as measured by the platelet adhesiveness and the clot retraction time.6 This suggested that changes in these properties of the platelets might play some role in the subsequent rapidly developing thrombocytopenia. Furthermore, a su...

A syndrome of platelet-release abnormality and mild hemophilia.

Two families were studied because of a hemorrhagic tendency. The presumptive diagnosis of von Willebrand’s disease was suggested by low factor VIII levels (7.5%33%), prolonged template Ivy bleeding time (9.5-17 mm), low platelet adhesiveness (0%-8%), normal platelet factor 3, and normal clot retraction. Further studies, however, showed abnormal platelet aggregation with ADP, epinephrine, and co...

Platelet functions in patients with meningococcal meningitis at the Kenyatta National Hospital, Nairobi.

OBJECTIVE To determine platelet abnormalities in patients with menigococcal meningitis. DESIGN Case control study. SUBJECTS Fifty seven cases of mennigococcal meningitis based on a cerebrospinal fluid gram stain for gram negative diplococcus or positive culture were recruited. Fifty-seven controls matched for age and sex were also recruited. The following platelet functions tests were perfo...

Effect in vitro on platelet function of two compounds developed from the pyrimido-pyrimidines.

VK 774 and VK 744, two new compounds developed from the pyrimido-pyrimidines, have been found to be powerful inhibitors of platelet function tested in vitro. They inhibit adenosine diphosphate (ADP)-induced platelet aggregation, and the release of platelet factor 3 by kaolin, and VK 774 also reduces platelet adhesiveness and inhibits platelet aggregation (;snowstorm' effect) in the Chandler tub...

Platelet functional abnormalities: a report of two familial defects in interaction between collagen and platelets and ADP release.

Two families have been studied, members of which have a lifelong hemorrhagic diathesis. In both families, a platelet functional abnormality has been found, with the coagulation mechanism intact. Findings common to both families were a prolonged bleeding time, reduced platelet adhesiveness, absence of the second wave of aggregation with ADP and reduced ADP release from platelets. The clot retrac...
